Mutations | K/R |
Epitope Location | K8R |
HXB2 Location | K43R |
Mutation Type | OV: observed variant RCR: replicative capacity reduced SF: susceptible form |
Method | CD8 T-cell Elispot - IFNy, Longitudinal study, Other, Relative replication capacity assay |
Note | Mutation K43R appeared at 91d post-screening, and was fixed by 175d post-screening by which time it was predominant (83%), indicating strong selection. No T-cell- or Ab-responses were detected against the mutant, and it was not a cryptic epitope, showing host-specific, non-adaptive immune selection pressures. The mutant was shown to be 12% less fit than the T/F virus over 3 comparative fitness passages until it was almost completely replaced by the T/F. |
