Note KkWIILGLNK is a late escape variant of KRWIILGLNK at anchor position-2 that reduces VRC substantially and abrogates binding to HLA-B*27. Clade B R264K though associated with progression to AIDS requires compensatory mutation S173A in order for the virus to survive. This mutation invariably follows the less potent but escaping variant, L268X. In Clade C, R264K does not follow L268X and has compensatory mutation S165N. Other compensatory mutations to C clade R264K are found between Protease-217 or Pro-225, PVHAGPIAP, in the cyclophilin A-binding loop.

Variant R264T does not require compensatory mutation S173A in order for the virus to survive and is typically selected without a preexisting L268X. A rare pediatric elite controller had maternal transmission of wild-type KK10 and mounted a strong response until selection of the R264T mutant which resulted in loss of KK10-response and progression to AIDS.
