• TEPITOPE algorithm was used to screen the most conserved regions of clade B consensus sequence for multiple HLA-DR binding epitopes. Peptides predicted to bind 16 or more HLA-DR molecules were synthesized and further assessed in binding assays and for T-cell recognition in a cohort of 32 Brazilian HIV-1 chronically infected individuals.
• While VLEWRFDSRLAFHHV is the reacting peptide, shorter WRFDSRLAF peptide was predicted by TEPITOPE to bind HLA-DR molecules. Flanking amino acids were added to increase the efficiency of in vitro peptide presentation to CD4 T cells.

• Co-administration of pGM-CSF (plasmid-encoded granulocyte-macrophage colony-stimulating factor) with a previously tested DNA vaccine encoding conserved, multiple HLA-DR binding HIV-1 subtype B peptides elicited broad, poly-functional and long-lived CD4+ T-cell responses. pGM-CSF co-immunization improved quality of HTL response - proliferation as well as IL-2, TNF-α and IFN-γ production.
• In addition to pGM-CSF co-immunization enhancing pooled HIVBr18-peptide-induced HTL responses, CTL proliferation and mono-functional CTL cytokine production was increased. HTL peptide Nef_180-194, VLEWRFDSRLAFHHV, ELISpot result increased upon co-administration of the vaccine with pGM-CSF.
