• CD4+ T cell responses were measured in a cohort of C-clade infected chronic, untreated subjects from Durban South Africa. Intracellular cytokine staining detected IFN-γ responses in 261/373 subjects, with the Gag responders having a significantly lower viral load and higher CD4 count than those with no detectable Gag response. Helper epitopes were determined by separate EliSpot screening on 32 subjects, using peptides spanning the entire clade C consensus sequence.
• 33 CD4+ peptides were targeted by 18/32 patients, with 27/33 peptides located in Gag region. 15/18 subjects targeted Gag, indicating a relative immunodominance of Gag. The magnitude of response did not correlate with viral load, but breadth of response had strong inverse correlation with viral load.
• SRIGILRQRRARNGASRS was targeted by 2/32 C-clade subjects.
