• Proliferative and cytokine (IFNgamma and Il-2) immune responses to 11 previously defined (Wilson 2001), and immunodominant and broadly cross-reactive Th epitopes and to p24 overlapping peptides were characterized in 10 LTNPs (chronically infected, asymptomatic, long term non-progressors, CD4+ T cells >500 after 15 years) and 7 SPs (slow progressors, CD4+ T cell declined to <500 after 15 years. Patients were treatment-naive.
• Gag-specific CD4+ T cells in LTNPs showed increased numbers of IFN gamma and IL-2 producing cells compared to SPs. Cytokine production and proliferative responses were negatively correlated with the viral load and positively correlated with the CD4+ T cell count. IL-2, but not IFNgamma, levels were correlated with proliferation.

• An engineered artificial polyepitope (polyE) vaccine consisting 50 CTL epitopes across Env, Gag, Pol, Nef and Tat, 7 helper epitopes, additional signal sequences and selected to cover the most of 35 HLA was delivered to BALB/c mice over a triple immunization schedule. Splenocytes from vaccinated mice were challenged by 6 CTL epitopes to determine magnitude of response.
• Vaccine epitopes were selected from the list at HIV Immunology database according to conservation across clades A, B, C. PolyE was linked to N-terminal ubiquitin (proteasomal trageting, MHC I loading), N-signal peptide (ER and secretory pathway delivery) and/or C-terminal LAMP-1 protein (directs immunogen from secretory pathway to lysosomes, MHC II loading) to give 3 constructs, viz. TCI-N (polyE), TCI-N2 (ER signal_polyE_LAMP-1), TCI-N3 (Ubi_polyE). All groups of mice developed both CTL and helper T-cell responses, but N-terminal ubiquitination mediated the highest expression of IL-2 and IFN-γ.
• Conserved HIV-1 epitope KTAVQMAVFIHNFKR was included in the vaccine and elicited T-cell responses.
