Notes

Dendritic cell mediated immunization of DNA plasmid constructs was studied in BALB/c (H-2d) and HLA-DR4 (DRA1*0101, DRB1*0401) transgenic mice. Most peptide-specific T-cell responses in the HLA-DR4 transgenic mice corresponded to human HLA-DR4 epitopes listed in HIV Immunology Database.
Gag peptide IYKRWIILGLNKIVR elicited CD4 T-cell responses in HLA-DR4 (DRB1*0401) transgenic mice immunized with pLAMP/gag-based prime and boost formulations.

The study measured CD4+ T-cell responses against epitopes in Gag p17 and p24 and concurrent endogenous plasma HIV-1 RNA epitope sequence variation. No intrapatient protein sequence variation was found in identified epitopes, indicating that escape from CD4-positive T-cell responses is not a common process in vivo.
2/22 patients responded to this peptide.

47 computationally selected helper epitopes from Gag, Nef, Env, Pol, Tat, were found to induce HIV-1-specific T-cell responses in a 38-member cohort of subjects infected with 10 different HIV-1 subtypes. 13 subjects were not taking any treatment, and HLA restriction was to multiple -DR and -DQ alleles. The peptides identified were therefore able to provide immune coverage for both viral and genetic variations across diverse ethnic backgrounds and as expected, viral load (VL) correlated inversely to the breadth and polyfunctionality of HTL response to these peptides.
Gag peptide IYKRWIILGLNKIVR is predicted to interact with 10 HLA-DR/DQ alleles. 5/38 (13.2%) of subjects had a CD4+ response to Gag-IR15.
