• Shu et al. [Biomed. Research International. Vol:2014, Article ID 272950] assembled a database that is comprised of sequences from 821 Chinese HIV isolates and 46 human leukocyte antigen (HLA) DR alleles identified in Chinese population. They then predicted 20 potential HIV epitopes using bioinformatics approaches, the combination of which within a polypeptide vaccine had a theoretical coverage of 98.1% of the population. In this study the vaccine was tested in transgenic mouse models and 6/20 epitopes found to elicit immune response in congenic mice as well as 5/20 in A2/DRB1 transgenic mice.
• Epitope GLIYSKKRQEILDLW was found in 6% Chinese population and 67% of DR+ Chinese. Though it did not induce EliSpot response in CB6F1 mice, significant response was seen in splenocytes from HLA-DRB1 transgenic mice.

• 6/20 epitopes selected bioinformatically for a recombinant polyepitope CD4+ T cell HIV vaccine, HIV20Th, elicited a response from CD4+ enriched PBMCs in at least 15% of 46 HIV-infected patients but all 20 epitopes elicited a response in at least one patient. 5/20 epitopes elicited a response in HIV20Th-immunized Balb/c mice. While a positive correlation was found between magnitude of the T-cell response and the amount of epitope bound to MHC II in HIV20Th-immunized Balb/c splenocytes, no correlation was found to in vitro peptide digestion patterns, binding affinities, or peptide-MHC dissociation assays. Half of the HIV20Th epitopes were included in a modified recombinant polyepitope CD4+ T cell HIV vaccine, HIV20Th-2, which elicited a response for all 10/10 epitopes in immunized Balb/c mice suggesting that in vivo competition for MHC molecules may limit the breadth of induced T-cell responses.
• GLIYSKKRQEILDLW (Nef 117) stimulated a CD4+ IFNγ EliSpot response in about 11% of 46 HIV-infected patients and HIV20Th-2-immunized Balb/c mice but not in HIV20Th-immunized Balb/c mice.
• Nef 117 bound MHC II with low affinity relative to the 20 peptides included in HIV20Th.
• Nef 117-overlapping peptides (KKRQEILDLWGP and YSKKRQEIL) were found presented on MHC II in HIV20Th-immunized immunized Balb/c splenocytes.tes.
