To study whether HIV-1 reverse transcriptase (RT) can be used as immunotherapeutically to prevent drug resistance, wild type and multidrug-resistant RT (RT1.14) were fused to lysosome-associated membrane protein (LAMP-1) and injected into BALB/c mice for assessment for subsequent immune response. Both chimeras induced mixed Th1/Th2 immune responses and generated cross-reactions between wild-type and drug resistant variants. Th1 responses included increased IL-2 secretion to RT-LAMP and RT1.14-LAMP, increased IFN-γ to RT1.14-LAMP and decreased TNF-α production to RT-LAMP.
Epitope specificity was altered with respect to IFN-γ as epitopes that induced its production were located in non-resistance-mutation regions of the RT protein. Immunization with RT1.14-LAMP fusion induced IFN-γ response against peptide FAIKKKDSTKWR and mutant peptide FAIKKKnnTgWR.
