Dendritic cell mediated immunization of DNA plasmid constructs was studied in BALB/c (H-2d) and HLA-DR4 (DRA1*0101, DRB1*0401) transgenic mice. Most peptide-specific T-cell responses in the HLA-DR4 transgenic mice corresponded to human HLA-DR4 epitopes listed in HIV Immunology Database.
Gag peptide, ASRELERFAVNPGLL, elicited CD4 T-cell responses in HLA-DR4 (DRB1*0401) transgenic mice immunized with pLAMP/gag-based prime and boost formulations.

CD4 T-cell responses in 36 HIV-1 positive individuals in different disease stages were mapped by IFN-γ EliSpot using a panel of 410 B-consensus peptides spanning all HIV proteins. Virus specific CD4+ T-cell responses (median=7, range of magnitude=990) were detected in 30/36 patients.
Gag and Nef responses dominated the CD4+ T-cell EliSpot responses in the study. No correlation between plasma viremia and the frequency or magnitude of HIV-1-specific responses was observed.
This peptide was recognized by 36% of the study group.
The peptides that were recognized by the most people were able to bind broadly to multiple HLA-DR molecules. This peptide showed high cross-reactive binding capacity and bound to 6/8 common HLA-DR molecules.
The most HIV-1 specific CD4+ T-cell responses were observed in acutely infected patients with treatment of primary infection who stopped after 1-3 treatments (acute STI), compared to patients continuing therapy. People with therapy during chronic infection had low responses; higher numbers of responses were seen among untreated people. 2/9 LTNPs responded to many peptides, comparable to acute STI.

67 Gag and Nef peptides restricted by HLA-DRB1 were characterized immunologically in 17 controllers and 9 progressors. HLA allele DRB1*15:02 was associated with low HIV-1 viremia and could promiscuously present peptides with lower avidity. HLA DRB1*03:01, however, was associated with high viremia and specific presentation of peptides. Moreover, Nef peptides were exclusively restricted by HLA-DRB1 variants linked with low viremia. Individuals with low viremia-linked HLA-DRB1 alleles also had greater breadth of HIV-specific CD4+ T cell responses. DRB1 allelic expression can impact HIV replication.
Overlapping peptide-6 (OLP-6), ASRELERFAVNPGLL, was promiscuous as it was restricted by 3 different HLA-DR across the HIV proteome as determined by IFN-γ EliSpot.

93 treatment-naive subjects in various stages of HIV-1 infection were analyzed for CD4 T-cell responses to the HIV B clade proteome using 410 overlapping peptides (OLPs). HTL responses were detected in 92% patients and breadth of these Gag responses were inversely correlated to viral load (VL). Targeting of 3 immunodominant Gag peptides was found to be linked to control of HIV infection.
Gag/Env mean response ratios were associated with effective immunity, and a robust marker of viral control, the lowest rations being found in elite controllers and the highest in high-VL progressors.
Immunodominant Gag peptide ASRELERFAVNPGLL was associated with spontaneous control, independent of the variation in HLA haplotypes. 18% of subjects responded to this peptide.

The cumulative contribution of Env-specific CD4+ T cells to viral control is seen here in 55 acutely infected individuals by longitudinally assessing them from acute to chronic infection. Highly targeted helper epitopes in acute infection were recognized at the same frequency in chronic infection. Gag specific-helper responses in acute responses were correlated with a lower viral set point during chronic infection, while the opposite was true for Env-specific resposnes. Subjects whose immunodominant responses were Gag-specific over Env-specific remained off ART for longer. Overall, helper responses are beneficial during acute HIV infection, and do not support disease progression, while these CD4+ responses remain stable in breadth, magnitude, dominance from acute to chronic infection.
ASRELERFAVNPGLL, Peptide 6, was the third most frequently targeted epitope tested and continued being targeted till chronic infection in some subjects.
