Notes

• HIV-specific CTL against non-escape epitope targets (HST-NEETs) can detect multiple, conserved, non-escaped epitopes and their variants. These HST-NEETS were generated in donors (either HIV+ or HIV-) against non-escaping epitopes found in PepMix (a mix of 15-mers from bivalent Gag and Pol epitopes called tHIVconsX), the entire Nef library of epitopes and known conserved epitopes in natural infection. When tested in vitro, HST-NEETs were able to, at clinical levels, lyse HIV-antigen presenting cells as well as produce polyfunctional cytokines with a pro-inflammatory response (TNF-α, IFN-γ). "Bivalent" mosaic peptides i.e. peptides with a variant were recgonized by HST-NEETs but not HSTs. Since HSTs usually decrease in patients on ART, HST-NEET infusions could be one method of eliminating HIV if it is reactivated out of latency.
• Peptide MOS-3, QMVHQAISPRTLNAW, a "bivalent" mosaic epitope from tHIVconsvX and its variant MOS-202, QMVHQALSPRTLNAW, were recognized by HST-NEETpos cell products from HIV+-Donor4.
• HST products from Donor4 could recognize the overlapping peptide GAG-38, GQMVHQAISPRTLNA.
