CTL response frequencies, breadths and polyfunctionalities (of IFN-γ, IL-2, TNF-α) were compared for recognition of consensus A and D HIV-1 infections using overlapping peptides (OLP) from consensus group M (ConM) peptides by EliSpot in 24 infected subjects. Comparable proportions of ConA, ConD and ConM peptides were recgonized when testing Gag or Nef OLPs, as they were for HIV-specific CTL or helper responses. Thus ConM OLPs may be used as reagents for clades A1 or D infected populations, though they are limited for evaluation of response breadth.
OLP GKLDAWEKIRLRPGGKKKYRL was consensus in ConM and ConA and elicited a strong CTL response in EliSpot, in 100% patients tested, 2250 sfu/10⁶ pbmc.
