• Mice were vaccinated and boosted with DNA vaccines containing Env gp 120 and a fusion of Gag-Pol-Nef. Epitopes in Pol and Env were mapped from reactive peptide pools.
• Fine mapping of a reactive 13-mer in Pol resulted in a positive EliSpot response from YYDPSKDLI.

Notes

• Mice were vaccinated with a construct consisting of the HIVconsv polyepitope plus the H epitope (RGPGRAFVTI), with the same construct given both as an adenovirus immunization and an MVA boost. Eight peptides elicited responses, 5 of which contained known epitopes. The remaining 3 peptides are considered to contain novel epitopes.
• Peptide 112 (YYDPSKDLIAEIQYW) was one of 8 peptides that elicited a response in vaccinated mice. It contained novel epitope YYDPSKDLI, revealed through minimal epitope mapping.
