Notes

The study compared B*27/*57-positive versus negative patients among a cohort of long-term nonprogressors and elite controllers with B*27/B*57-negative progressors. Overlapping 15-mers that elicited responses in multiple patients, or were among the strongest responses in a single patient, were narrowed down by truncation to define optimal epitopes, 11 of which were novel.
YSPTSILDI was recognized by 1/1 LTNP/EC, and 1/1 had it as a dominant response. The restricting HLA (Cw*01) was confirmed experimentally.

Polymorphic epitope regions can be targets for cross-clade CTL response. In addition to 6 conserved AE epitopes, 9 epitopes polymorphic between clades AE and B were identified in chronically infected Japanese subjects. This is the first comprehensive study to identify cross-clade CTL by using overlapping HIV-1 peptides.
There were no significant differences in total magnitude of CTL response between 26 clade A/E-infected and 401 clade B-infected subjects when tested with 11-mers from HIV-B Nef, Gag and Pol. 6 epitopes were novel viz. Gag LL11, Pol SV9, Pol GI9, Gag RV8, Gag AP8 and Gag EG11.
Clade B epitope Gag p24 YI9, YSPTSILDI, was found in 1032/1644 Clade B and 30/315 Clade A/E Los Alamos database HIV-1 sequences. It was found in 2/26 A/E-infected subjects of the KI cohort. CTL induced by this clade B peptide were able to recognize clades B peptides and A/E peptides evenly.
CTL from A/E-infected subjects stimulated by the consensus B YI9 peptide used cross-recognized both clades B and A/E infected cells.

Specific T cells - from 120 ART-naive, infected Japanese subjects previously exposed to the tHIVconsvX vaccine - that have the ability to suppress HIV-1 replication were identified as recognizing 17 optimal Gag epitopes. 9/17 eptiopes were associated with lower pVL and higher CD4 count, of which 5 remain associated with positive clinical outcome in 221 HIV-1 infected subjects and cross-recognized major HIV-1 variants. 2/5 epitopes were novel. Candidate tHIVconsvX vaccine is composed of 6 conserved Gag and Pol regions.
Cw*0102-restricted epitope YSPTSILDI was found in 15-mer peptide pairs C070/071 and C072/073. 6 HLA-carrying responders to this epitope were detected by EliSpot out of 50 subjects.

Notes

In this original study showing the protective role of HLA-C restricted CTL responses in nonwhite Chinese HIV-positive subjects, the authors find an association between a single nucleotide polymorphism (SNP) upstream of HLA-C and immune pressure on the infecting Clade B HIV. This mutation is found to correlate with enhanced cell surface HLA-C expression, lower VL, greater maintenance of CD4 cells resulting in slower progression to AIDS and now also a greater IFN-γ production by HLA-C-restricted CTLs.
The SNP is -35CC and is associated with 29% mutations in viral DNA as opposed to only 4% mutations in -35TT subjects. These mutations that were associated with the SNP in HLA-C were denoted C-mut. 6 C-mut were found in proviral DNA - 3 in previously known epitopes and 3 in peptides containing as yet unpublished epitopes.
For Gag epitope YSPTSILDI, C-mut is due to a T280V substitution that decreased recognition.
