Notes

• HLA-B*5801 is associated with slow progression. Six B*5801-positive subtype C-infected subjects were followed for up to 31 months from infection. CTL responses during acute infection predominantly targeted the TW10 and KF9 epitopes. Transmitted escape and compensatory mutations appeared to limit the protective effect of the B*5801 allele.
• Longitudinal sequencing from the 6 patients revealed variants that were assayed for EliSpot responses. Escape mutations (WPnVRERm, WPdVRERm, WPeVRERm, WPdVgERm, WPnVREgm, WPdVREkm) in this epitope arose in 1/6 individuals.
• In EliSpot assays of overlapping peptides across the entire proteome, 1/6 patients targeted this epitope, with confirmed single-peptide response.
