• Fifteen peptides were identified from NL4-3-infected C*12:02 single-allele-transfected cells, revealing preferences for A (strongest), S, E, L, I, and V at the 2nd position and F, Y, L, and K at the C terminus position. One novel peptide RAIEAQQHL and 2 previously reported peptides (ILKEPVHGVYY and VARELHPEY) are immunodominant and were the only 3 peptides of 13 assessed that elicited a CTL response in 20 chronically HIV-1-infected C*12:02+ (a protective allele) subjects.
• VTNPATIM was observed in HLA-peptide complexes from C*12:02-expressing cells infected with NL4-3.
• VTNPATIM sequence was observed in more than 5/20 chronically HIV-1-infected Japanese C*12:02+ subjects, but despite measurable affinity for C*12:02, a VTNPATIM-specific CTL response was not detected in the same cohort.
• Other experiments in this study were completed using the corresponding clade B consensus sequence VTNsATIM or cohort-specific variants (see record 62514). VTNPATIM is contained within overlapping peptide MSQVTNPATIM which was also studied in this paper.

• 107 HLA-A*02-restricted epitopes of varying lengths (novel, nested or overlapping) were discovered by using mass spectroscopy to identify peptides bound to HLA on 3 kinds of HIV-infected cells (66 from 293T cells, 20 from a B cell line, B-MDC, and 39 from primary CD4+ T cells). 80-91% of peptides partly overlapped known reported epitopes by >5 aa, while 5-20% were novel. Eluted peptides tended to cluster in nested groups.
• Majority of peptides isolated were from Gag and Pol, while Gag peptides from p15 outnumbered those from the more immunogenic p24. 19/23 peptides were tested and were immunogenic. Cytosolic processing and degradation was confirmed.
• Highly immunogenic novel Gag-p15 epitope VTNPATIM was naturally processed and eluted from live B-MDC-HIV and CD4+ T cells. This peptide has an extended N-terminal as compared to previously known epitopes.
