• Two model HXB2-derived DNA immunogens, protease (PR) and reverse transcriptase (RT), were administered to Balb/c mice by intramuscular (IM) or intradermal (ID) single immunizations with subsequent electroporation revealing that ID delivery increased the production of IFNγ and IL-2 in PR-immunized, but not RT-immunized, splenocytes. A robust antibody response was only observed in RT-immunized mice and was significantly enhanced with ID, vs. IM, delivery. Epitopes PQVTLWQRPLVTIKI, AIGTVLVGPTPVNII, and KVVPLTNTTNQK also elicited in vitro responses in immunized splenocytes but it is less clear if this was predominantly CTL or CD4+ T cell-mediated recognition.
• After ID immunization with a plasmid containing PR, ˜7% of reactive CTLs (the most robust CTL response of the assessed PR peptides) and ˜1% of CD4+ T cells were VLVGPTPVNII-specific and mostly produced IFNγ alone or IFNγ+TNFα.
• Compared to IM, immunization via ID delivery and subsequent in vitro stimulation produced significantly more VLVGPTPVNII-splenocytes secreting IFNγ and IL-2.
