Cross-clade vaccine recognition was studied using either single-, multi or several anatomically separated single-clade vaccines. Limited cross-clade response was seen with single-clade administrations. Multi-clade vaccines gave immune interference (antagonism and original antigenic sin), reducing CTL response. Simultaneously administered but anatomically separated vaccines from clades A, B, C decreased antagonism and increased immune responses.
After immunization with Clade A vaccine containing Epitope E, SYHRLRDFI, T cells were able to respond to epitope variant SYHRLRDFv, as well as variants SYHRLRDci, SYHRLRDll and SYHRLRDli at <50% magnitude. Variants SYrhLRDFI, iYHhLRDll and SYrlLRDll were not recognized.

An engineered artificial polyepitope (polyE) vaccine consisting 50 CTL epitopes across Env, Gag, Pol, Nef and Tat, 7 helper epitopes, additional signal sequences and selected to cover the most of 35 HLA was delivered to BALB/c mice over a triple immunization schedule. Splenocytes from vaccinated mice were challenged by 6 CTL epitopes to determine magnitude of response.
Vaccine epitopes were selected from the list at HIV Immunology database according to conservation across clades A, B, C. PolyE was linked to N-terminal ubiquitin (proteasomal trageting, MHC I loading), N-signal peptide (ER and secretory pathway delivery) and/or C-terminal LAMP-1 protein (directs immunogen from secretory pathway to lysosomes, MHC II loading) to give 3 constructs: TCI-N (polyE), TCI-N2 (ER signal_polyE_LAMP-1), and TCI-N3 (Ubi_polyE). All groups of mice developed both CTL and helper T-cell responses, but N-terminal ubiquitination mediated the highest expression of IL-2 and IFN-γ.
Conserved HIV-1 epitope SYHRLRDFI restricted by mouse BALB/c HLA-H-2^d was included in the vaccine and elicited T-cell responses.
