CD8 T-cell responses were examined in mother-infant pairs. Escape variants were commonly detected in maternal plasma. Early infant plasma viruses showed heterogeneity of gag and nef gene sequences as well as mother-to-child transmission of CD8 T-cell escape variants. The stability of escape mutants in the infant over time was determined by infant HLA haplotype and viral fitness.
Sequential plasma specimens from infants showed changes in CD8 T-cell epitope sequences, suggesting that infants are capable of generating virus-specific CD8 T-cell responses.
The mother was A*02- (M-1004) and carried a variant form of the epitope, MThNPPIPV, which she passed to her A*02+ child (P-1115). This form persisted in her child for 12 months.

107 HLA-A*02-restricted epitopes of varying lengths (novel, nested or overlapping) were discovered by using mass spectroscopy to identify peptides bound to HLA on 3 kinds of HIV-infected cells (66 from 293T cells, 20 from a B cell line, B-MDC, and 39 from primary CD4+ T cells). 80-91% of peptides partly overlapped known reported epitopes by >5 aa, while 5-20% were novel. Eluted peptides tended to cluster in nested groups.
Majority of peptides isolated were from Gag and Pol, while Gag peptides from p15 outnumbered those from the more immunogenic p24. 19/23 peptides were tested and were immunogenic. Cytosolic processing and degradation was confirmed.
Immunogenic Gag-p24 epitope MTNNPPIPV was naturally processed and eluted from live 293T and B-MDC-HIV cells.

HLA-A11-restricted CTL epitopes in 2 male Han Chinese subjects (one ART naive and one receiving HAART) were determined. A novel epitope was discovered and 5 other putative epitopes confirmed using 425 overlapping peptides spanning the entire HIV-1 clade B proteome. By employing intracellular cytokine staining, the authors developed a new method of mapping optimal epitopes and defining their HLA restriction efficiently. In spite of its being the most common HLA type in China, A*11-restricted epitopes were recognized less frequently than other epitopes, and A*11-restricted responses formed a mean of only 13% of the responses against HIV.
This epitope, HPVHAGPIAPGQMREPR, was detected within overlapping peptideS LHPVHAGPIAPGQMREPR, LKETINEEAAEWDRLHPV and AAEWDRLHPVHAGPIA.

Subtype-specific substitutions in two HIV Gag regions (fragments F1, amino acids (aa) 5-46, and F2 aa 100-143) result in novel adaptations that affect epitope processing efficiency. Consequent low epitope abundance is associated with weak T cell response and greater immune escape and therefore with disease state. Conversely, epitopes from these regions when restricted by certain rare HLA I alleles, are associated with greater production and resulting CTL response, therefore delayed disease progression.
Most epitopes were found to be processed as precursors with N-terminal extensions beyond the optimal epitope. Only 4 epitopes in this region were associated with delayed progression - HLA-B*5701/03-restricted IW9 (ISPRTLNAW), KF11 (KAFSPEVIPMF), TW10 (TSTLQEQIGW) and HLA-B*2705-restricted KK10 (KRWIILGLNK).
Regardless of ethnic group, white or African, a significant three-way interaction was found between HLA allelic frequency, epitope yield and ethnicity for epitopes of N-terminal fragment F1 (NF1) and C-terminal as well as middle F2 (CF2 and MF2). C-terminal F1 (CF1) epitope yields however, showed a significant difference between ethnic groups. Also, for identical HLA-mediated selection pressure in both Africans and whites, HIV-C had a lower concentration than HIV-B, especially at lower HLA allelic frequencies.
Epitope MTNNPPIPV, MV-9, is from fragment NF2-MF2, and restricted by HLA-A*02. 50% MTNNPPIPVG, MV-9's subtype B, natural processed epitope form, accumulated after 4 hours processing by constitutive proteasomes, and 82% with immuno proteasomal processing. For subtype C, the natural processed form was not given.

This is a trial of a multi-epitope DNA vaccine that included 21 CTL epitopes, which are conserved among clades A,B,C,D and CRFs and are supertype binding peptides of A1, A3 and B7 supertypes. Participants were from US and Botswana.
The vaccine was safe and well-tolerated, but the rate of CTL induction was low. CD8+ responses (and no CD4+ responses) were detected in 3/26 subjects using chromium release assay and in 2/42 subjects using IFN-γ ELISPOT assay.
This epitope was included in the vaccine.

DNA vaccine EP HIV-1090 evaluation in patients receiving ART was done. This vaccine is composed of multiple CTL epitopes, all associated with certain HLA supertypes. Differences of CTL response rate in vaccine and placebo recipients however, was not significant.
MTNNPPIPV is a Nef epitope encoded in the EP HIV-1090 polyepitope vaccine.

A placebo controlled trial was run in 36 HIV-1-uninfected adults, where either a DNA vaccine or recombinant modified vaccinia virus Ankara vaccine (rMVA-mBN32) was administered. Both vaccines encoded the same 21 CTL and 18 HTL epitopes but did not elicit strong CTL or helper responses as compared to vaccines containing entire open reading frames of HIV proteins.
CTL response as γ-IFN and IL-2 production was detected in 3/25 subjects who were vaccinated. PBMCs from these responders were tested with specific peptides and only one subject had a positive response to the epitope Gag 271, MTNNPPIPV. The entire list of CTL epitopes is found in this database in Gorse2008a.

21 supertype-A2, -A3 and -B7-restricted epitopes predicted to be immunogenic by HLA-peptide binding motifs and phenotypic frequency in individuals were recognized by HIV-1+ subjects in ELISpot assays and shown to bind with high affinity to minimum 3 HLA-alleles of the restricting supertype. These epitopes were included in DNA vaccine EP HIV-1090 that avoided immunodominant responses and did not restrict subject immunity to predicted HLA-type when tested in mice. Transgenic mice mounted a vaccine-response similar to that induced by ELISpot assay in humans.
Epitope MTNNPPIPV of the HLA-A2 supertype bound most strongly to HLA-A*6802, -A*0203 and -A*0601 and also to -A*0206 and -A*0202. It was conserved 89% in subtype B. 0/22 HLA-A2 supertype expressing HIV-1+ subjects mounted a positive ELISpot response to Gag271.

Long-term nonprogressors (LTNPs) had strong memory resting CD8+ T-cell responses against the majority of epitopes tested, (18 for the A2 supertype, 16 for the A3 supertype) while the effector cells of long-term nonprogressors recognized far fewer epitopes
Progressors had memory resting CD8+ T-cells that recognized far fewer epitopes than LTNPs.
A positive correlation between effector CD8+ T-cells and plasma viremia and a negative correlation between CD8+ effector T-cells and CD4+ T-cells was observed, which may contribute to the inability of LTNPs to clear virus.
This epitope can bind three of the five HLA-A2 supertypes alleles (A*0201, A*0202, A*0203, A*0206 and A*6802).
