• Previous studies of the HIVconsv vaccine containing a chimeric protein with alternating HIV-1 Clade A, B, C, D conserved regions showed that subdominant epitopes could induce strong CTL response correlating with Gag and Pol-specific IFN-γ production. Here the specificity i.e. the epitopes themselves were characterized as well as the functionality, (i.e. cytokine production) of CTL from 3 subjects of the HIV-CORE 002 trial.
• CTL elicited by HIVconsv from healthy human volunteer 411 (A*0201/B*0801, B*5101/Cw*0303, Cw*0701), when expanded upon stimulation with peptide Pol 80, GFRKYTAFTIPSINN (Pol 277-293), gave a strongly hierarchical immune response. It contained the mapped epitope KYTAFTIPSI.

Notes

• A novel CTL clone was defined with a panel of recombinant vaccinia-RT-infected B-LCL target cells using PBMCs donated by a patient who was HIV-seropositive for 6 years and had not received any antiretroviral therapy.
• There is evidence that some CTL epitopes are poorly presented on the surface of infected cells, but this RT epitope was recognized as effectively on HIV-infected cells as on peptide-pulsed targets.

• Since CTL response in natural HIV-1 infections is mostly against variable but immunodominant epitopes, lymphocytes from 12 vaccine(HIVconsv)-exposed subjects of the HIV-CORE 002 study were used to determine and define 14 less variable but typically subdominant, optimal novel epitopes.
• Epitope KYTAFTIPSI (KI10) is within peptide GFRKYTAFTIPSINN (HC080) that was recognized by VID411, 1/12 vaccinated subjects. KI10 is predicted to be restricted by HLA-Cw*0701.
