• Immunodominant responses of 25 uninfected, healthy subjects to highly conserved regions in HIV-1 were induced against an Ad35 vaccine containing a subtype A fusion protein of Gag-RT-Int-Nef-Env. Virus inhibition assay (VIA) confirmed cross-reactivity to 7 other HIV-1 isolates including subtype B (IIIB, CH77, CH106, ELI), subtype A (U455), and subtype C (97ZA012, 247FV2). CTL responses were to a median of 4 epitopes per vaccine, directed mostly against p17. Peptide mapping and patient HLA were used to define optimal epitopes.
• Reactive peptide KGSPAIFQSSM, induced immune response as determined by EliSpot and ICS in expanded CTL from uninfected, vaccinated subject 040 (HLA-A*0201, B*0702, B*5801, C*0702, C*0718). It has previously been associated with HLA-B*07 as optimal epitope, SPAIFQSSM.
