Notes

• 2 novel B*5201-restricted and 1 novel Cw*1202-restricted epitopes were determined that contained 4 previously identified HLA-B*5201-associated polymorphisms inversely correlated with plasma viral load (pVL). The epitope variants were found to be selected by B*5201 or Cw*1202 CTL and affected replication competence of the virus.
• Novel immunodominant epitope IY11, ILKEPVHGVYY, was isolated, containing HLA-Cw*1202-associated polymorphism, Pol 466. 19/80 B*5201-Cw*1202 subjects responded to this epitope.
• The K466R mutation was found to be IY11-specific CTL-selected, and an individual K466R-mutant NL4-3 virus affected viral fitness by decreasing replication competence. Replication competence was much lower for multiple mutant viruses with four mutations, either NL4-3-T68A-K466R-T993S-I935L or NL4-3-T68I-K466R-T993S-I935L.

• HLA-B*5201-C*1202 haplotype which has strong linkage disequilibrium and is found in approximately 20% Japanese, is protective during HIV-1 infection by suppression of HIV-1 replication via CTL and NK cells. Cw*1202 is known to work with KIR2DL2 in NK-cell mediated control.
• Several immunodominant HLA-Cw*1202-restricted HIV-1 epitopes were previously discovered. One of them was Pol IY11 (ILKEPVHGVYY). IY11-specific T cells were detected in 20% HLA-Cw*1202+ chronically infected HIV-1 subjects. Infected responders to IY11 challenge had a significantly lower pVL (plasma viral load) and higher CD4+ T-cell count, mediated via HLA-Cw*1202.

• Fifteen peptides were identified from NL4-3-infected C*12:02 single-allele-transfected cells, revealing preferences for A (strongest), S, E, L, I, and V at the 2nd position and F, Y, L, and K at the C terminus position. One novel peptide RAIEAQQHL and 2 previously reported peptides (ILKEPVHGVYY and VARELHPEY) are immunodominant and were the only 3 peptides of 13 assessed that elicited a CTL response in 20 chronically HIV-1-infected C*12:02+ (a protective allele) subjects.
• ILKEPVHGVYY, a known C*12:02-restricted immunodominant epitope, was observed in HLA-peptide complexes from C*12:02-expressing cells infected with NL4-3 and is identical to corresponding clade B consensus.
• ILKEPVHGVYY-specific CTL responses were detected in 5/20 chronically HIV-1-infected C*12:02+ Japanese subjects.
• ILKEPVHGVYY demonstrated C*12:02 binding with a peptide/HLA refolding assay but not with a HLA stabilization assay suggesting low affinity.
