Notes

Cross-clade vaccine recognition was studied using either single-, multi or several anatomically separated single-clade vaccines. Limited cross-clade response was seen with single-clade administrations. Multi-clade vaccines gave immune interference (antagonism and original antigenic sin), reducing CTL response. Simultaneously administered but anatomically separated vaccines from clades A, B, C decreased antagonism and increased immune responses.
After immunization with A-clade vaccine, index epitope P, IFQSSMTKI, and variant IFQaSMTKI were recognized. Variant IFQcSMTKI was not recognized.

Using a candidate vaccine immunogen (HIVA), immunodominant epitopes were sequentially removed to test whether subdominant responses could be protective. Subdominant epitopes induced robust responses and became immunodominant in the absence of higher-ranked epitopes. Thus vaccines focused on less-variable regions with subdominant epitopes could be effective.
When the immunodominant epitope H (RGPGRAFVTI) was deleted, subdominant epitope P (IFQSSMTKI) became immunodominant.
Mutation of epitope P to disrupt the anchor residues caused subdominant epitope G1 (AMQMLKDTI) to become dominant. The mutant used to replace epitope P (IFgSSMTKa) was confirmed to lack binding to MHCs H-2D^d, H-2K^d and H-2L^d.
Fine mapping of this epitope, P, revealed IFQSSMTKI to be the optimal epitope.

The HIVA polyprotein contains p24, p17, and a polyepitope string of conserved epitopes that were selected to have particularly good cross-reactive potential for the A-clade epidemic in Kenya [Hanke et al., Nat. Med., 6:951-955 (2000)]. A candidate vaccine was constructed with Mycobacterium bovis bacillus Calmette-Guerin (BCG) as the vector.
Mouse response to subdominant epitope P increased with increasing dose of vaccine, but peaked much lower than the immunodominant epitope H.

HIV clade C gag, pol and fusion gagpol vaccines were compared in mice. Breadth of T cell responses was improved in mice immunized with gagpol fusion genes, compared to single antigen constructs. 5 new murine CD8+ T cell epitopes were mapped.

An engineered artificial polyepitope (polyE) vaccine consisting 50 CTL epitopes across Env, Gag, Pol, Nef and Tat, 7 helper epitopes, additional signal sequences and selected to cover the most of 35 HLA was delivered to BALB/c mice over a triple immunization schedule. Splenocytes from vaccinated mice were challenged by 6 CTL epitopes to determine magnitude of response.
Vaccine epitopes were selected from the list at HIV Immunology database according to conservation across clades A, B, C. PolyE was linked to N-terminal ubiquitin (proteasomal trageting, MHC I loading), N-signal peptide (ER and secretory pathway delivery) and/or C-terminal LAMP-1 protein (directs immunogen from secretory pathway to lysosomes, MHC II loading) to give 3 constructs: TCI-N (polyE), TCI-N2 (ER signal_polyE_LAMP-1), and TCI-N3 (Ubi_polyE). All groups of mice developed both CTL and helper T-cell responses, but N-terminal ubiquitination mediated the highest expression of IL-2 and IFN-γ.
Conserved HIV-1 epitope IFQSSMTKI restricted by mouse BALB/c HLA-H-2^d was included in the vaccine and elicited T-cell responses.

A novel ovine adenoviral vector (OAdV) was compared to human AdV5 vector for the delivery of HIV clade A vaccine in mice. The ovine vector primed T cells more efficiently, but was less persistent than AdV5.
Subdominant epitope IFQSSMTKI was used to assess the effectiveness of the vaccines.
