• RV144 vaccine was administered and T-cell based sieve analyses applied to compare differences between HIV-1 sequences of vaccinated versus placebo, infected recipients. A sieve effect i.e. binding escapes was seen only for 3 CTL epitopes - GTIKGGEMK (GK9), TSIGDKMQK (TK9), KMQKEYALL (KL9) - all of subtype B, MN strain within the V2 loop of rgp120 immunogen and particularly at residue 169 in the V2 loop (hypothesis is that these MN-derived subtype B vaccine epitopes applied cross-reactive pressure on the infecting CRF01_AE subtype epitopes).
• Novel A*11-restricted CTL epitope KL9, KMQKEYALL, was discovered.
• Vaccine efficacy was greater in A*02+ than in A*02- subjects.
• GK9 HLA binding escapes were significantly greater in vaccine recipients.
• Breakthrough sequences from the CRF01_AE cohort containing variant epitopes are reported in GenBank, accession# JX446645-JX448316.
