Notes

A metric to determine degree of HIV adaptation to HLA and HLA profile was developed. It was found that transmission of pre-adapted HIV to recipients with the associated HLA in a population resulted in elevated VL, impaired immunity and CD4+ T cell decline. Also, pre-adaptation of circulating HIV accounts for much of the variation in immune outcome that was previously attributed to HLA alleles.
Transmission was studied in HIV-C chronically-infected subjects from Southern African cohorts. Adapted FKL9 (FPVkPQVPL) when transmitted to PBMCs elicited a depressed CTL response in an in vitro killing assay as compared to non-adapted FL9 (FPVRPQVPL) when infecting similar PBMC targets.

Notes

An 11-member cohort of acutely infected patients was studied to find that CTL cross-recognition of variant epitopes during acute infection is absent or poor as opposed to during chronic HIV-infection.
Nef epitope FPVKPQVPL, FKL9_68-76,-specific T cells were less cytotoxic to variants than autologous FKL9 in acute infection. No variants tested cross-reacted with T cells specific to autologous FKL9.

The ability of T cells from early infected subjects to recognize Nef clade B HIV-1 epitope variants was tested using both a consensus peptide panel and a standardized potential T-cell epitope panel (PTE-B). The magnitude and breadth of PTE-optimized peptide panels was significantly higher than CON peptides'. 29 potential epitopic domains were detected in Nef using the PTE peptide set.
A PTE-B peptide, EEEVGFPVkPQVPLR containing the epitope FPVRPQVPLR (FL10) was found in two subjects and elicited an IFN-gamma immune response.
HLA-B35 and -B07 restriction for FL9 were presumed based on the subject's having the HLA allele and publication in the Los Alamos database.

The extent to which set-point viremia is affected by early HIV-infection T cell responses to either conserved or variable epitopes was studied. 23 ART-naive subjects were characterized for CTL responses for 6 months, postinfection. While most CTL responses (73/123) were targeted against variable HIV-1 epitopes, recognition of conserved epitopes (16/123) was associated with lower set point viremia. This was independent of HLA type and also functional avidity of the CTL response. Most of the conserved responses were targeted to Gag and Pol epitopes.
FPVKPQVPL, restricted by B35 or B7, was recognized by 2 patients with the restricting allele.
