• A long term non-progressor (LTNP), Subject 15160, was studied for detailed analyses of CTL response to epitopes correlated to the subject's viral quasispecies. Epitope mapping revealed CTL response to 6 epitopes in p17, p24, RT, Env and Nef. Results show that (a) multiple epitopes were targeted simultaneously (b) viral heterogeneity/antigenic diversity is not necessary for such a multispecific CTL response (c) LTNP infection does not need to be associated with unique epitope targeting as 5/6 epitopes targeted have been seen in other subjects with disease progression (d) HIV-1-specific CTL can be highly active, disallowing viral escapes, but broadly cross-reactive (recognizing known variants) even with low viral load (VL).
• A Gag-specific clone, CA1, from the LTNP subject 15160 recognized a p24, HLA-B*14-restricted novel 10 aa. epitope, DRFYKTLRAE, which overlaps previously identified RAEQASQEV. However, 9 aa. DRFWKTLRA is the optimal epitope.

• The immunodominant CTL response in a long-term survivor was to this highly conserved and functionally relevant epitope.
• By testing mutations in an HXB2 background, it was found that all mutations within the epitope that abrogated CTL recognition also abolished viral infectivity.
• The epitope in this study overlaps the major homology region for which highly conserved residues exist in all known lenti- and onco-viruses and yeast transposons.

• Receptor 2B4 (CD244) is a member of the SLAM family of receptors that regulate lymphocyte development and function. 2B4 can activate or inhibit immune responses, and its expression on T cells increases during the course of HIV infection. In this study, responses to 2B4+ and 2B4- T cell populations were compared using HIV negative HLA B14 and B27 donors. CD8+ T cells were stimulated with HLA B14- and B27-restricted epitopes. Secretion of IFN-γ by HIV epitopes was lower than after stimulation with influenza epitope, and expression of signaling adaptor SAP was downregulated in 2B4+ CD8+ T cells upon HIV peptide stimulation. Thus 2B4+ CD8+ T cells may contribute to poor viral control of HIV.
• Previously-documented epitope DRFYKTLRAE, and its escape variant DRFYKiLRAE, were among epitopes used for comparisons.
