Notes

To study immune correlations with natural HIV-2 control, a cohort in Guinea-Bissau was studied for 2 decades. 33 controllers were significantly associated with a CTL response that was high-magnitude and polyfunctional. The CTL response was of restricted breadth as it was to a specific epitope for the most part, as opposed to similar HIV-1 associated CTL - 80% of the controllers mounted a response to only one HIV-2 epitope. There was no correlation with perforin upregulation. Gag-specific, but not Env-specific responses defined viral control.
HLA-B*14-restricted immunodominant HIV-2 p26 epitope, DRFYKSLRA, may contribute to viral control as seen by EliSpot.

CTL lymphopoietic potential and virus suppressive capacity were studied in 20 cART-naive, HIV-2-positive subjects with controlled viremia (HIC2). Production of HIV-2 Gag-specific CTL with (1) early memory differentiation as well as (2) robust effector potential was preserved in HIC2.
2/20 subjects were B*1401-positive and all recognized Gag p24 epitope DRFYKSLRA (DA9).

Bioinformatic analysis of capsid sequences identified fewer codons under positive selection & greater purifying selection in HIV-2, vs. HIV-1, suggesting increased constraints on evolution may result in persistence of HIV-2-specific CTL responses. Unlike HIV-1, the 2 HIV-2 codons under positive selection are not within any known CTL epitopes, but flank 3/4 known capsid epitopes. Five HLA-associated HIV-2 polymorphisms were detected and all were within or flanking known or predicted CTL epitopes.
Capsid epitope DRFYKSLRA, known to be B^*14-restricted, was conserved in 96.4% of 86 ART-naive HIV-2-mono-infected subjects.
All 4 B^*14-positive subjects had positive EliSpot responses to DRFYKSLRA (215-1051 SFU/10⁶ PBMCs) and had conserved dominant autologous epitope sequences.
Codon 313, which is 5 AA downstream of DRFYKSLRA, was shown to be under significant positive selection. Codon A313 & polymorphism A313P were respectively positively & negatively associated with HLA allele B^*5801 and are located in predicted epitope QTDAAVKNW (or QTDpAVKNW) which is likely restricted to B^*5801.
Codon P294 & polymorphism P294S were respectively positively & negatively associated with HLA allele C^*0401. Codon 294 is located 6 AA upstream of DRFYKSLRA & 3 AA upstream of predicted epitope SYVDRFYKSLRA wihch is likely restricted to C^*0401.
