• Allogenic HIV-specific A*02-restricted CTL were generated from HIV-naive adults (HNA-T) or cord blood (CB-T) with intent to use for adoptive T cell therapy meant to control HIV rebound post-hematopoietic stem cell transplant (HSCT). A good manufacturing practice (GMP)-grade protocol was used to generate the CTL which were shown to be functionally able (in vitro cytolysis and HIV suppression), non-exhausted, and broadly recognising of multi-antigen epitopes in Gag, Nef, and Pol.
• Mapping using pool matrices and EliSpot were used to determine peptides recognized by CTL generated from HIV-negative donors (adults or cord blood). Pol DDCVASRQDED was not recognized by any CTL from HIV-negative HLA-A*02+ donors but by CTL from one HIV-positive adult-derived donor.
