Notes

• Crystal structure of the Pol283-8 (TAFTIPSI, TI8) epitope with HLA-B*5201 shows that presentation is similar to Pol283-8:HLA-B*5101, ensuring Pol 283-8 cross-presentation by both alleles. Chronically infected Japanese subjects carrying HLAs B*5101 or B*5201 and Pol 283-8-specific CTL suppressed HIV-1, though more strongly with B*5101 than B*5201.
• In Japanese and Caucasian cohorts, the RT I135T mutant was associated with HLA-B*5101, and RT I135X mutants were weakly associated with HLA-B*5201. Viral suppression of I135X mutant HIV-1 was possible by Pol 283-8-specific, B*5101-restricted CTL but not by Pol 283-8-specific, B*5201-restricted CTL.
• Lower TCR affinity of Pol 283-8:B*5201 as compared to TCR affinity for Pol 283-8:B*5101 is probably due to a conformational difference in the N-terminal region of Pol 283-3.
