• A metric to determine degree of HIV adaptation to HLA and HLA profile was developed. It was found that transmission of pre-adapted HIV to recipients with the associated HLA in a population resulted in elevated VL, impaired immunity and CD4+ T cell decline. Also, pre-adaptation of circulating HIV accounts for much of the variation in immune outcome that was previously attributed to HLA alleles.
• Transmission was studied in HIV-C chronically-infected subjects from Southern African cohorts. Non-adapted QK9 (QIYAGIKVK) when transmitted to PBMCs elicited a much greater CTL response in an in vitro killing assay than did adapted QPK9 (QIYPGIKVK) when infecting similar PBMC targets.

• Rolland2016 is a comment on Carlson2016 [Carlson et al. Nat. Med. 22: 606-613 (2016)] which suggests that there are "holes" in the host immune repertoire resulting in a deterioration of immune response to HLA-adapted CTL escape mutations that are transmitted. Extent of HLA-adaptation of the infecting HIV-1 increased over the first 2 years particularly in subjects with high viral load (VL). Such adapted epitopes elicited less CTL response and this phenomenon was accentuated among transmission pairs who shared HLA. While the Carlson et al. study included no Env data, it does suggest that vaccine strategies should focus on conserved elements of HIV-1.
