The study showed that some epitope mutations, particular those at anchor residues, can abrogate binding between HLA and killer Ig-like receptor (KIR). Thus some epitope variants could affect immune recognition by affecting KIR-HLA interactions.
Several amino acid changes in IW9 interfered with HLA-B*57 binding with KIR, particularly mutations at the -1 (most notably to E,K,D,L,R) and -2 (most notably to D,K,L) anchor positions.

The study showed that some epitope mutations, particular those at anchor residues, can abrogate binding between HLA and killer Ig-like receptor (KIR). Thus some epitope variants could affect immune recognition by affecting KIR-HLA interactions.
In two HIV-infected patients, TW10 mutants G9E and T3N appeared early in infection. The T3N change abrogated T cell recognition of the TW10 epitope, but G9E mutation, which occurred earlier in infection, had only a modest effect. The T3N variant disappeared by day 350, but the G9E mutant increased steadily.
The KIR3DL1 carried by these patients is known to recognize HLA-B*5703 and is associated with good viral control. G9E variant severely reduced interaction between KIR3DL1 and HLA-B*5703. In addition, mutations at the -1 (most notably to E,K,D,L,Q,R) and -2 (most notably to D,K,L,P) anchor positions tended to interfere with binding.
