B*27:05+ subjects had significantly higher peptide-MHC binding avidity than B*27:02+ subjects as determined by luminescent oxygen channeling immunoassay (LOCI).
• Structural modeling with a modified Gag KK10 peptide with W substituted for C-terminal residue (PC) confirmed that 268L in B*27:05 constrains bulky PC residues. It is known that B*27 requires R at position 2 in the binding peptide which corresponds to 264R for Gag KK10. Consistent with immunodominance, Gag KK10 residues 264R and 268L (HXB2) were shown to be under strong selective pressure in B*27:05-positive subjects compared to B*27-negative subjects.

• Peptide NYTPGPGVRYPLTFGWCF was the immmunodominant EliSpot response out of 410 OLPs spanning the B clade proteasome in 7 B*27:02+ patients. The optimal 9mer epitope VRYPLTFGW (Nef VW9) was defined.
• 100% of 7 B*27:02+ subjects responded to the immunodominant epitope VRYPLTFGW (Nef VW9) as compared to about 21% of 19 B*27:05+ subjects. Nef VW9-B*27:02 avidity was significantly higher than Nef VW9-B*27:05 as determined by luminescent oxygen channeling immunoassay (LOCI). Structural modeling with a modified Gag KK10 peptide (W substituted for C-terminal residue or PC) confirmed that, unlike B*27:05, B*27:02-specific polymorphism L268A permits the bulky PC residue W in Nef VW9.
• Consistent with immunodominance, Nef VW9 residues 137L and 139F (HXB2) were shown to be under strong selective pressure n B*27:02+ subjects compared to B*27-negative subjects.
