10E3: A polyepitope vaccine was designed based on a recombinant GST fusion protein containing three repeats of the 2F5 core epitope ELDKWA combined with the V3 region peptide GPGRAFY. The genetic variability of the neutralizing epitope GPGRAFY was studied and its distribution in different subtypes was assessed. The dominant motifs of the epitope were unequally distributed in different HIV-1 subtypes, and it was shown to be one amino acid shorter in the majority of HIV-1 group O-strains compared to the group-M strains. HGD65: This MAb was isolated from memory B cells of HIV-1 infected donor using an improved EBV immortalization method combined with a broad screening strategy. HGD65 bound to Env proteins from clades B, C and F. It cross-competed with MAb HGA9 for binding to gp120. HGD65 neutralized 6/20 isolates in the HOS-based assay, 3 Tier 1 and 3 Tier 2 viruses belonging to clades B, AD and AG. In TZM-bl assay, HGI75 neutralized 7/46 isolates, 5 Tier 1 and 2 Tier 2 isolates belonging to clades B and AG.
