N49P7: Extensive structural analysis of N49P6, and comparison with similar N49P7, defines a novel class of near-pan-neutralizing CD4bs bNAbs (N49 series). Relative to N49P6, N49P7 has a slightly higher neutralization potency (IC80 values < 50 μg/mL for 98.3% of 117 pseduoviruses in a multi-tier global panel vs. 83.8%) due to CDR polymorphisms (53M, 56Q, & 57V in CDRH2, lack of R insertion in CDRH3, and 91Y in CDRL3). N49P7: Monoclonal antibodies, including some that are near-pan-neutralizing, were extracted directly from the plasma of two elite neutralizers (N60 & N49). Against a multi-tier multi-clade 117 pseudovirus panel, extremely potent and broad bNAb N49P7 individually recapitulated 100% of subject N49 anti-gp120 polyclonal plasma IgG antiviral breadth. X-ray crystallography of bNAb N49P7 suggested a unique ability to bypass the CD4bs Phe43 cavity, while reaching deep into highly conserved residues of Layer 3 of the gp120 inner domain. Author provided supplementary unpublished neutralization assay data (IC80 values and additional pseudoviruses) that can be accessed via the LANL HIV database CATNAP tool.
