5B11: This study reported the development of a CD4-independent, CCR5-tropic variant of the pathogenic molecular clone SIVmac239, termed iMac239. This could help to evaluate SIV pathogenesis and host immune responses in the absence of the immunomodulatory effects of CD4+ T cell targeting and infection. In neutralization assay iMac239 was resistant to anti-CD4BS murine mAb 5B11. Swanstrom2016 (SIV)
5B11: In order to generate bnMAbs to SIV,two immunization protocols were compared using different preparations of oligomeric SIV envelope (Env) glycoproteins.In the first protocol, mice were immunized with soluble gp140 (sgp140) from CP-MAC variants of SIVmacBK28, SIVsm B670 CL3 and SIVsm543-3E. The Abs isolated were against V1/V2 loop and V3 loop. In the second protocol, mice were immunized with unfixed CP-MAC-infected cells and MAbs were screened for the ability to inhibit cell-cell fusion. Five classes of neutralizing MAbs were identified. 5B11 interfered with CD4 binding site.In oligomer sensitivity assay, 5B11 was more reactive with the oligomer-containing Env than with the monomeric gp120 preparation.
