2C6: The antigenic structure of Gag-Env pseudovirions was characterized and it was shown that these particles can recapitulate native HIV virion epitope structures. The Gag-Env pseudovirions were further used to identify a subset of antigen-specific B cells in chronically infected HIV subjects. The newly discovered Ab 2C6 showed homology in its CDR3 region with MAb 47e. 2C6 showed significant binding only to the Gag-Env pseudovirions and not to the gp120 monomer. 2C6 showed low-level neutralization activity against HIV BaL which increased with the addition of sCD4. It is suggested that 2C6 binding epitope is a quaternary structure on the gp120/41 trimeric Env spike.
