AB#31: This scFv Ab was generated from a phage-displayed Ab library generated from PBMCs from a seropositive donor and selected by biopanning with gp160. It recognized a conformational epitope on gp41 and it showed high affinity binding in the nanomolar range. Binding of AB#31 was inhibited with the donor serum, indicating presence of Abs with similar specificity in this patient. The epitope of AB#31 was also recognized by a broad range of patient sera: 21/22 sera from North America and 20/20 sera from African patients all inhibited binding of this Ab. deHaard1998 (antibody binding site, antibody generation, variant cross-reactivity, kinetics, binding affinity)
AB31: GPI-anchored and secretory scFvs of AB31 were generated. GPI-scFvs were localized in the lipid raft of the plasma membrane. Cells transduced with either the secretory, or the GPI-anchored AB31 scFvs, did not show neutralization breadth and potency against 11 tested pseudotype viruses belonging to clades A, B, B', C and E, nor against 6 wild type HIV-1 strains. Ab 31: The Ig usage for variable heavy chain of this Ab was as follows: IGHV:1-46*01, IGHD:2-2, D-RF:1, IGHJ:3. Non-V3 mAbs preferentially used the VH1-69 gene segment. In contrast to V3 mAbs, these non-V3 mAbs used several VH4 gene segments and the D3-9 gene segment. Similarly to the V3 mAbs, the non-V3 mAbs used the VH3 gene family in a reduced manner.
