• 8H3:7 non-neutralizing, linear epitope-binding murine mAbs induced by group M consensus Env, CON-6 immunization were characterized. 8H3 bound to the first constant region, C1 at the N-terminus (aa 30-42) of Env, part of the inner domain of gp120 Env. Native decosylation of HIV JRFL increases 8H3 binding which is predicted to bind an N-linked glycosylation site. It binds Hep-2 cells cytoplasmically. 8H3 has 92% cross reactivity with Env from clade A, and 100% with clades B, C, CRF01_AE and CRF0X_BC. • 8H3: This MAb was cloned from mouse hybridoma after immunization of the animals with subtype C Env protein. 8H3 cross-reacted with a subset of different subtype Envs in ELISA, but did not recognize HXB2 or SIVcpzUS Envs. 8H3 did not neutralize any of the tier 1 and tier 2 subtype B and C viruses. This suggests that the conserved epitope targeted by this Ab is not accessible on the functional Env proteins. Gao2009 (antibody generation, neutralization, variant cross-reactivity)
