COMMENT SIV-MAC239 was derived from SIV-MAC251. In this study, two juvenile
Rhesus macaques were injected with the supernatant of cell cultures
which had been infected with the SIV-MAC239 molecular clone.
Mm243-86 was injected with 1 milliliter of the supernatant of a
41-day culture of HuT78 cells, which had 25,592 CPM of RT activity.
Mm326-87 was injected with 1 milliliter of supernatant from Rhesus
macaque PBMC transfected for 18 days with SIV-MAC239, which had
100,000 cpm of RT activity.
As a control, to assess the variability of the virus in the culture
supernatant which was injected into the macaques, the 41-day
culture supernatant which was injected into Mm243-86 was used to
infect another culture of fresh Hut78 cells and cultured for an
additional 31 to 55 days. 5 clones were sequenced from this
secondary culture. The CTL72 in the name of this clone, indicates
that it is one of the control clones, taken at 72 days of total
culturing (41 in the initial culture and 31 in the secondary).
